ABSTRACT

Objective:Based on network pharmacology to study the mechanism of Shaoyao-Gancao Decoction in treating tension-type headache. Methods:Searched for the active ingredients and potential targets of Shaoyao-Gancao Decoction from TCMSP database, and adopted the targets of tension-type headache from GeneCards, DisGeNET, Drugbank and OMIM databases. Then obtained all the intersections of Shaoyao-Gancao Decoction and tension-type headache, and uploaded them to the STRING databases to construct a PPI network and conduct topological properties analysis. Finally, established a Chinese medicine regulatory network of Chinese medicine-components-target genes-disease by Cytoscape 3.6.1 software. To perform the GO function enrichment analysis and KEGG analysis on the core targets. Results:There were 51 intersections of Shaoyao-Gancao Decoction and tension-type headache. The topological properties analysis suggested that CASP3, JUN, HSP90AA1, MAPK1, STAT3, CCND1, ESR1, RELA, PTGS2, MAPK14 may be the potential targets for the treatment of tension-type headache in Shaoyao-Gancao Decoction. Gene ontology enrichment analysis showed 876 biological processes, 101 molecular functions and 62 cellular components. KEGG pathway enrichment analysis showed 25 related signaling pathways, including TNF signaling pathway, serotonergic synapse, NOD-like receptor signaling pathway, Dopaminergic synapse and Sphingolipid signaling pathway. Conclusion:The treatment of tension-type headache by Shaoyao-Gancao Decoction verified the characteristics of multi-component, multi-target and multi-pathway, which provided reference for the clinical medication.

ABSTRACT

Objective To explore the clinical effect and safety of methimazole combined with 131I in treatment of hyperthyroidism.Methods Patients (83 cases) with hyperthyroidism accepted in Xi'an Jiaotong University Affiliated San Er Ling Yi Hospital from June 2012 to June 2015 were selected and divided into observation group with 42 cases and control group with 41 cases.Patients in observation group were given methimazole combined with 131I,and patients in control group were given PTU combined with 131I.Then the clinical effect,thyroid gland serological indexes,adverse reations,and reoccurrence rates of two groups were observed and compared.Results The total clinical efficacy of observation group was 95.24%,which was obviously higher than 70.73% of control group with statistically significance (P < 0.05).TSH was getting higher after treatment of two groups,and T3,T4,FT3,FT4,TRAb,and TPOAb were getting lower after treatment of two groups.And TSH in the observation group was significantly higher than that in the control group,T3,T4,FT3,and FT4 were significantly lower than those in the control group (P < 0.05).The adverse reaction rate of observation group (14.29%) was obviously lower than that of control group (46.34%) with statistically significance (P < 0.05).The reoccurrence rate of observation group (4.76%) was obviously lower than that of control group (26.83%) with statistically significance(P < 0.05).Conclusion Methimazole combined with 131I has good effect and safety in treatment of hyperthyroidism,which is worth of clinical application.

ABSTRACT

Objectives To investigate characterization of imipenem resistance among imipenem non susceptible Pseudomonas aeruginosa isolated from patients who treated without imipenem and explore risk factors of imipenem resistance.Methods From April,2006 to March,2008,a total of 37 non-susceptible to imipenem without imipenem therapy isolates were collected from affiliated 3201st Hospital of Medical College of Xi'an Jiaotong University.The minimum inhibition concentration (MIC) to 11 antimicrobial agents were determined by the broth dilution method.We also tested imipenem MIC combined with efflux pump inhibitor PAβN.PCR was performed to check for the presence of carbapenem-hydrolylzing MBL genes and oprD gene.The expression level of oprD2 and ampC were evaluated by qRT-PCR.Molecular typing was performed using PFGE.Results There is significant difference ( t =- 2.9004,P < 0.01 ) of the average number days of therapy between with two or more antibiotics in the 16 patients (20.0 ± 9.5 ) d and that with only one antibiotic in the other 21 patients ( 12.6 ± 4.4 ) d before imipenem-non-susceptible strains were isolated.In all 37 strains,32 strains showed resistance to more than three antibiotics.The MBL gene ( IMP-9 ) was only found in one strain,but its phenotype is negative,oprD2 gene from the 29 strains were found forward inserted by ISpa1328.Thirty-five isolated were considered to have no oprD expression.The patterns of the total DNA of 37 strains appeared six PFGE types.The 26 strains belonged to C2 PFGE type.In the presence of PAβN,all 37 strains increased sensitivity to meropenem.Conclusion Fluoroquinolones and cephalosporins treatment could play an important role in imipenem non-susceptible production in the research isolates.
